JPMorgan expects the phase three success to support Chinese mRNA cancer vaccine development and demand for specialized CXO services, with benefits concentrated among companies with differentiated pipelines and existing relevant service capabilities.
What exactly did the phase three success validate?
Moderna’s individualized mRNA cancer vaccine, intismeran autogene, combined with pembrolizumab, met both the primary endpoint of recurrence-free survival (RFS) and the key secondary endpoint of distant metastasis-free survival (DMFS) in a phase three trial of adjuvant treatment for melanoma after surgery. JPMorgan believes this provides important clinical validation for the use of mRNA vaccines in cancer beyond infectious diseases.
The trial enrolled 1,137 patients whose tumors had been completely resected, with 757 in the combination group and 380 in the monotherapy group. The RFS hazard ratio was 0.58, p<0.001, close to the 0.561 reported at two years of follow-up in phase two; the DMFS hazard ratio was 0.62, p=0.002, showing statistically significant benefits from combination treatment in both recurrence and distant metastasis.
The efficacy came with some safety trade-off: grade three or higher adverse events occurred in 23.6% of the combination group, above the 20.3% in the monotherapy group. The bank considers the overall safety profile manageable.
Why did Moderna shares still fall 17.6% despite the positive data?
Moderna shares fell 17.6%, while the biotech ETF XBI fell 0.75% over the same period. JPMorgan attributed the decline to high expectations and investor debate over the scope for expanding indications.
The main shortfall was in the distant metastasis measure: at a median follow-up of 24.6 months in phase three, the DMFS hazard ratio was 0.62; in phase two, it was 0.35 at a comparable follow-up of approximately 23 months and 0.41 at five years. A lower hazard ratio implies a greater relative benefit, and DMFS is generally regarded as a stronger surrogate for overall survival than RFS, so this gap affected investors’ assessment of the magnitude of efficacy.
Some clients also questioned the appeal of adjuvant treatment alone because another phase two trial showed that pembrolizumab given before and after surgery was superior to its use only after surgery. The bank noted, however, that the trials used different measures, EFS and RFS, and that the treatment design of Moderna’s trial had long been public.
Where can Chinese CXOs meet the resulting demand?
JPMorgan expects clinical validation to support incremental demand for specialized services including mRNA research and development, process development, manufacturing, plasmid and template supply, in vitro transcribed mRNA, and translational oncology research. The link to potential benefits lies in companies’ existing mRNA capabilities and positions across the industry chain.
Specifically, WuXi AppTec’s TIDES business handles mRNA manufacturing, while its testing and biology businesses cover early research; WuXi Biologics provides research and development capabilities through WuXi Vaccine; and GenScript provides in vitro transcribed mRNA, plasmids, templates and related services.
How far can confidence in Chinese vaccine pipelines improve?
The bank expects confidence to rise in differentiated, individualized or off-the-shelf mRNA cancer vaccine pipelines in China. Examples include Akeso’s phase one AK154 and RGL-270 from Regenelead, a subsidiary of Hengrui. For the latter, data disclosed at ASCO in 2026 showed an RFS rate of 100% at 18 months among 16 patients with resectable pancreatic ductal adenocarcinoma.
Similar technologies have also seen failures: BioNTech’s BNT113 recently missed the progression-free survival endpoint in a phase two/three trial. JPMorgan therefore emphasizes that indication selection, trial design and vaccine design determine the outcome of individual programs; the phase three success increases confidence in the technology approach.
