Why does the 41% response rate encourage Goldman Sachs?

  • GFH276 is an oral pan-RAS(ON) molecular glue. Clinical data in previously treated patients with RAS-mutant solid tumors have been disclosed for the first time. Among 61 patients with pancreatic ductal adenocarcinoma in the 40–160 mg dose groups, 25 achieved an objective response, giving a response rate of 41% and a disease control rate of 92%.
  • This result exceeds the thresholds of 30% and 20% that Goldman Sachs previously set for second-line and third-line or later monotherapy, respectively. Patient composition is also important: approximately 73% had received at least two prior lines of treatment, 78% had liver metastases, and 98.6% had received chemotherapy. The efficacy results came from a population predominantly treated across multiple prior lines.
  • As of 15 September 2026, this phase I dose-escalation trial had enrolled 98 patients, including 73 with pancreatic cancer, with a median follow-up of 18 weeks. Goldman Sachs gave a positive assessment on that basis, which remains data from an early dose-exploration stage.

How does it compare with similar drugs?

  • The second-line and third-line or later pancreatic cancer monotherapy response rates cited by Goldman Sachs were, respectively: 29% and 22% for daraxonrasib; 40% and 23% for ERAS-0015; and 39% and 24% for JAB-23E73. GFH276 achieved 41% in a mixed population predominantly treated in the third-line setting or later. The numerical comparison across trials is attractive, although the groups differed in their composition by treatment line.
  • There were also signs of efficacy in non-small cell lung cancer: among 11 patients in the 80–160 mg groups, the objective response rate was 46% and the disease control rate was 73%. Goldman Sachs also noted that the sample was small and follow-up short, making the data susceptible to fluctuations; pancreatic cancer therefore remains the firmer basis for assessment at present.

What does manageable tolerability mean in practice?

  • Among 80 patients in the 40–160 mg groups who did not receive preventive medication, there were no dose-limiting toxicities or grade 4–5 treatment-related adverse events. The incidence of grade 3 events was 27.5%, the dose reduction rate was 17.5%, and the discontinuation rate was 1.3%. The low discontinuation rate is an important reason Goldman Sachs considers tolerability manageable, although dose reductions were still needed during treatment.
  • Rash burden differed by cancer type: the incidence of any-grade and grade 3 rash in pancreatic cancer patients was 79% and 1.6%, respectively, below the overall cross-cancer rates of 84% and 7.5%. Management believes that more lung cancer patients had received immunotherapy, which may be associated with their higher rash burden.
  • The incidence of stomatitis was also substantial: the any-grade rate was 48%, close to daraxonrasib’s 53% and above ERAS-0015’s 19%. However, no grade 3 stomatitis or related treatment discontinuations were observed.

Why is dose selection, rather than further escalation, the next priority?

  • No clear increase in response rate with higher doses was observed between 40 and 160 mg, and steady-state drug exposure was similar at 120 mg and above. At 200 mg and above, dose-limiting toxicities occurred, primarily consisting of rash. Management believes toxicity is more dose-dependent than efficacy, so subsequent additional enrollment has focused on 40–160 mg.
  • Goldman Sachs therefore believes the recommended phase II dose may be around 80 mg. Accumulation of skin toxicity over longer treatment is an important constraint on dose selection. Management hopes to keep the long-term grade 3 rash rate in single digits and the grade 1–2 rate at 60%–70%, and to reduce incidence further through more proactive clinical management.
  • Goldman Sachs views the update as a positive step toward developing GFH276 into a pan-RAS backbone drug and will focus on recommended doses for monotherapy and combination regimens. The bank maintained its buy rating and set a 12-month target price of 52 Hong Kong dollars.
